Can I use a program such as Nessus to scan my own computer for vulnerabilities?

If not, what other ways can I go about detecting holes and patching them? I figure rather than having my system hacked into I'd like to learn how to prevent that from happening. I use a good firewall and anti-virus but I know that doesn't stop most intelligent hackers.
